  I have just started, but seem to have a mastery point that I cant use? Am I missing something?
  
  I go into the Codex and the mastery tab, and it says click here to try to upgrade the current ability, but nothing happens.

  Mastery system is currently disabled afaik. They wanted to see the baseline abilities in effect to be able to tune them better.
